Renin Angiotensin System and Its Therapeutic Approaches
نویسندگان
چکیده
The renin–angiotensin system (RAS) has an important role in the control of blood pressure, hypertension, diabetes, and in the regulation of salt and volume homeostasis of organisms as well as in the heart failure, and other related renal dysfunctions. Renin is the main protease enzyme of the RAS. In kidney, Juxtaglomerular (JG) cells are most important site for renin production. Renin act on Angiotensinogen (AOG) and cleave Angiotensin I and form different Angiotensins-ANG II, ANG 1-7, ANG 1-9 using Angiotensin converting enzyme (ACE) and Angiotensin receptors (AT-R). RAS inhibition using βBlockers, Angiotensin-Converting Enzyme Inhibitors (ACEIs), Angiotensin Receptor Blockers (ARBs), Direct Renin Inhibitors are proven important for the study of prevention of several renal diseases and cardiovascular diseases. Additionally, several drugs such as vitamin D receptor activators, endothelin antagonists, Aldosterone antagonists, or monocyte chemotactic protein-1 antagonists are also used in controlling blood pressure, endothelial dysfunction and heart failure. Aliskiren, Enalapril and other inhibitors has proved a valuable therapeutic approach in different diseases.

Roles of Renin-Angiotensin System in the Regulation of Prostate Cancer Bone Metastasis: A Critical Review
Mestastatic prostate cancer cells (MPCCs) frequently metastasize to bone, which is a “favorite soil” for colonization and proliferation of MPCCs. Prostate cancer bone mestastasis is tightly associated with tumor-induced bone lesions, most commonly caused from the etiological imbalance between osteoblastic bone formation and osteoclastic bone resorption, and from the anti-tumor immune response. ...
متن کاملRenin-angiotensin system and unilateral ureteral obstruction
Unilateral ureteral obstruction (UUO) is a clinical scenario that leads to obstructive nephropathy. UUO alters the expression of many mediators in the ipsilateral kidney. Renin-angiotensin system (RAS) is involved in UUO. Angiotensin II (Ang II) and angiotensin 1-7 (Ang 1-7) as the main arms of RAS influence kidney function which may alter by UUO. Ang II via Ang II receptor subtypes I (AT1R) ...
متن کاملResveratrol Suppresses Cardiac Renin Angiotensin System in the Late Phase of Left Ventricular Hypertrophy
Background and objectives: Resveratrol(3,5,4′-trihydroxy-trans-stilbene) is a natural polyphenole phytoalexin which exerts potential cardioprotective effects, but the cellular and molecular mechanisms responsible for these effects are still unknown. Cardiac renin angiotensin system (RAS) over-activation plays an important role in pathogenesis of left ventricula...
متن کاملI-34: New Management Options for Ovarian Hyperstimulation Syndrome
Ovarian Hyperstimulation Syndrome (OHSS) is a rare complication of super ovulation and in vitro fertilization (IVF). Although our understanding of its pathophysiology is incomplete, induction of development of multiple follicles, perturbation of the renin angiotensin system and exposure to luteinizing hormone (LH) or human chorionic gonadotropin (hCG) are essential contributing factors to its d...
متن کاملNEP, ACE and Homologues: The Pathophysiology of Membrane Metalloproteases
The zinc metalloprotease, neprilysin (NEP), plays a role in the metabolism of cardiovascular, inflammatory and neuropeptides, including mitogenic peptides such as bombesin. In the cardiovascular system, NEP has a primary role in the inactivation of natriuretic peptides but also contributes to local metabolism of angiotensin, endothelins and bradykinin. Hence NEP is seen as a potential therapeut...
